Lipogenesis in Particle-free Extracts of Rat Liver

چکیده

A particle-free extract has been prepared from rat liver which is capable of incorporating isotopic acetate, pyruvate, and acetyl CoAl into long chain fatty acids. While both cytoplasmic and mitochondrial enzymes are necessary for the complete system, evidence is presented suggesting that the presence of electrolyt,es in the homogenizing medium may act to extract the lipogenic enzymes of the mitochondria into the soluble part, iclefree phase. The coenzyme requirements of this system have been established. In addition, a marked stimulation by cupric ion and by several acids of the tricarboxylic acid cycle has been observed. Some evidence is presented as to the mode of action of the latter.

Changes in lipid synthesis in rat liver during development.

1. Lipogenesis, as measured by the incorporation of (14)C-labelled glucose or acetate into fatty acids in liver slices, is high in foetal and adult rat liver but is low in the liver of the suckling rat, especially with glucose as substrate. 2. The rate of synthesis of non-saponifiable lipids from glucose is about 15 times as great in the liver of the 18-day foetus as in adult liver. Activity in...

The immediate effects of insulin and fructose on the metabolism of the perfused liver. Changes in lipoprotein secretion, fatty acid oxidation and esterification, lipogenesis and carbohydrate metabolism.

1. When livers from fed rats were perfused with blood containing elevated concentrations of rat insulin or blood to which fructose was added, the oxidation of free fatty acids was depressed and their esterification was increased. 2. Raised concentrations of insulin or addition of fructose increased secretion of triglyceride in very-low-density lipoproteins, but only insulin caused more of the f...
